My favorite thing, that I am constantly amazed by, is how easily you can connect with someone from across the country that you otherwise would never have "met". I really have enjoyed some of the relationships that I have made via Twitter.
I think my least favorite thing is how hard it is to find people via their search feature.
My favorite thing is the speed of information I can get from Twitter. You can look at your twitter stream for 20 minutes or less and be able to catchup/know almost all the current breaking news in any sector (as long as you're following all the right people).
My least favorite thing is their search. It is hard to find specific things.

I have been a slow adopter of Twitter. I like being kept up to date on current events of those that I follow. However, what I like the most is that my favorite brand(s) have a customer service Twitter account. It seems like each time I reach out to a customer service department via Twitter, I get a quick response and greater service. That is what has stood out the most so far.

Very interesting question. Here is my conflict: I use Twitter relentlessly BUT I literally can not list one favorite thing about it. I use Tweetdeck so I barely even know what Twitter looks like.
Ok, so I will answer the question: best thing about Twitter for me is the access to information relevant to me.
Favorite - simple, well formed short-format.
Least favorite - the Fail Whale. I've been seeing that guy way too much lately.

Least favorite thing: spam bots
Most favorite: Cutting to the chase. Short and sweet.
Least favorite: Search - it's hard to find anything unless it's an exact match.
Favorite thing: Openness to connect with anybody
Least favorite thing: Difficulty in following related conversation threads. Hash tags are still crude (and BTW the spam sucks too).
Favorite thing: Twitter is an EXCELLENT way to find other people talking about or interested in exactly what you're looking for. It is also an unprecedented online platform for real-time human interaction, similar to Facebook, but also different in that it is far more open for all to see. Uber transparency!
Least Favorite thing: I agree that their Search is inadequate, but I think their major shortcoming is allllllll of the 3rd party apps that have popped up to provide other features and services to supplement Twitter. (eg. TweetDeck, HootSuite, Tweeple, there are zillions). It would be nice to have all (or most) of Twitter's potential functionality and applications on the platform itself.
My favorite this about Twitter is being able to interact with diverse people and groups. The ability to share information is also a plus.
Likes: the ability to connect with people I'd ordinarily never meet
Dislikes: the search functionality, the noise, and the spam. I'd like to be able to more easily filter for what I'm interested in.
I agree with Roger along with many other excellent answers. Twitter provides a quick way to get not only up to date information, but cutting edge and progressive insight from industry professionals. For "Dislikes" I'd say that yes, the spam can get annoying, but while the 140 character limit might seem like a disadvantage at first, it's actually an asset of this platform.
